Ndufs2 encodes a protein that contributes to the ubiquinone-binding site of mitochondrial complex I (MCI). Homozygous knock-out animals manifest clear, levodopa-responsive parkinsonism that is attributable to MCI dysfunction. In contrast to conventional Parkinson's disease (PD) models in which dopamine (DA) is depleted rapidly throughout the basal ganglia, the staging of pathology in these mice enables an assessment of how regional deficits in DA release are coupled to behavior.
